Organization Overview

We study nuclear pore complexes (NPCs), which provide the only known sites for nucleocytoplasmic exchange. Using a variety of techniques and yeast as a model system, we are determining the composition and fine structure of the NPC and relating this to its sites of interactions and reactions with soluble nuclear transport factors. We are also examining associations between NPCs and other components of the nuclear periphery. We hope to gain a better understanding of the part the NPC plays in nuclear regulation and maintenance.

Overview

The Laboratory Manager's duties will be divided across lab management / administrative and technical / scientific duties. Administrative duties will primarily involve general product ordering and lab organization (65% time). Scientific research duties will initially center around the preparation of antibodies against target proteins (35% time). The scientific focus will evolve as the project does.

Responsibilities

Lab management/administrative duties:

  • Place Orders (mainly for Rout lab, but also for Cross and Chait labs)

  • Pick up from Receiving / Stockroom and mailroom, and deals with issues involving receipt or non-receipt of items and payment

  • Responsible for organizing the following general lab upkeep, including:

  • Put away clean glassware and dishes around the sink

  • Keep stock of standard consumables and reorder as needed

  • Check CO2 and liquid N2 tanks; request new tanks from custodial

  • Fill main lab consumables (falcon tubes, syringes, serological pipettes) and autoclave as necessary

  • Fill 70% ethanol, bleach, Windex bottles

  • Autoclave milliQ water

  • Making stock solutions in lab, e.g. gel running buffers, transfer buffers, Coomassie stain

  • Check -80 freezer for temperature and ice build-up

  • Check LN2 storage

  • Check Disposal Bins (Biological waste & glass & foil & soft plastic)

  • Chemical waste - Fill out Cards & Drop off at Lab Safety.

  • Aid research projects (as described above)

  • Act as Lab Safety Officer: looking for safety violations (eyewash blocked, unlabeled waste, ethanol in cold room); organizing annual lab safety training updates for all lab members

  • Schedule Lab Meetings and sending reminders, orders food, sets up AV (including Zoom)

  • Assist in updating NCDIR website/Rout Lab Website/NCDIR Youtube/Rout Lab Twitter/NCDIR Twitter;

  • Perform autoclaving as needed

  • Perform related duties and responsibilities as assigned/requested

Scientific research duties

  • Prepare antibodies against target proteins, including polyclonal fractions and nanobodies, which will also be used in affinity capture screening and mass spectrometric studies, and involvement in those screening studies and downstream analyses.

Qualifications

Bachelor’s degree required; educational emphasis in Biology or Biochemistry preferred.

Skills required include standard biochemical / cell biological bench skills (solution preparation, protein expression and purification, cell and tissue culture, electrophoretic analyses) as well as antibody purification and immunoprecipitation. Molecular cloning experience a plus. Must be detail oriented, highly organized, and computer literate.

The salary of the finalist selected for this role will be set based on various factors, including but not limited to organizational budgets, qualifications, experience, education, licenses, specialty, and training. The hiring range provided represents The Rockefeller University's good faith and reasonable estimate of the range of possible compensation at the time of posting.
